Summary:
Many questions about mechanism of action, pharmacokinetics, and clinical applications of leucovorin rescue are being asked again. How does leucovorin rescue cells from high-dose MTX ? How much leucovorin should be given to protect normal tissue and maximize antitumor activity? In individual patients, how should leucovorin rescue be modified to maximize therapeutic index? Some answers to these quastions are provided in this review.
